降纤酶对大鼠急性心肌梗死后梗死周边区心室肌细胞动作电位和L型钙电流的影响

摘    要:目的 探讨降纤酶对大鼠急性心肌梗死后梗死周边区心室肌细胞动作电位和 L型钙电流的影响。方法 制成大鼠急性心肌梗死模型, 分两组, 一组对照, 另一组腹腔注射降纤酶0. 2 U/ (100g·d), 共 7 d, 酶解分离梗死周边区心室肌细胞, 采用全细胞膜片钳技术记录其动作电位和 ICa,L。结果 降纤酶组梗死周边区心室肌细胞 ERP/APD90增大, 受抑制的 ICa,L有所恢复, 峰电流密度显著增大, I V曲线下移, 失活曲线右移, 失活后再恢复速度加快。结论 降纤酶使急性心肌梗死后梗死周边区心室肌细胞ERP/APD90增大, ICa,L有所恢复, 因此具有减少急性心肌梗死后室性心律失常和改善心功能的作用。

Abstract:Objective To investigate the effects of Defibrase on action potential and L-type calcium current of ventricular myocytes in peri-infarcted area after acute myocardial infarction in rats.Methods The rat model of acute myocardial infarction was made.Defibrace (0.2 U/100 g every day) was injected for 7 days.Single ventricular myocytes were isolated enzymatically from the zone adjacent to infarcted area and the similar region in the control hearts.Action potentials and L-type calcium currents were recorded by whole-cell patch clamp techniques.Results In Defibrase group,ERP/APD 90 was increased,peak density of I Ca,L was significantly improved,steady-state inactivation curve was shifted to the right,and I Ca,L recovered more fast.Conclusion Defibrase increases ERP/APD 90 and peak density of I Ca,L,suggesting it can decrease the occurrence of arrhythmia after myocardial infarction and improve cardiac function.
