Abstract:Objective To investigate the effect and molecular mechanism of CTRP9 on reducing the activation and migration of cardiac fibroblasts(CFs). Methods The cell proliferation activity of CFs were detected by MTS kit. The expression of α-SMA was detected by immunofluorescence staining and western blotting to evaluate CFs-to-myofibroblast transformation. Wound healing assay and transwell test were used to test the migration ability of CFs. Western blotting was used to detect collagenⅠ, Ⅲ and CTGF. Results CTRP9 significantly inhibited the increase of TGF-β1-induced CFs proliferation(P<0.001), transformation to myofibroblasts(P<0.01), but enhanced migration ability(P<0.001), and the expression of collagen I, collagen III, and CTGF(P<0.001). PKA inhibitor H-89 blocked those inhibitions of CTRP9(P<0.001, P≤0.05, P<0.001, P<0.001). Conclusion CTRP9 reduces TGF-β1-induced proliferation, transformation to myofibroblasts, migration and the expression of fibrosis-related proteins of cardiac fibroblasts via PKA to exert anti-fibrosis effects.
