安体舒通对2型糖尿病大鼠肾脏的保护作用及对肾组织mcp-1表达的影响

摘    要:目的:观察安体舒通对糖尿病大鼠肾脏组织的保护作用及其对尿单核细胞趋化蛋白-1(UMCP-1)排泄和肾组织mcp-1表达的影响。方法:将30只SD大鼠随机分为模型组(20只)和对照组(10只),再将模型组随机分为糖尿病和治疗组,于给药后第8周检测2型糖尿病大鼠及安体舒通治疗大鼠正常对照大鼠的尾外周血糖和糖化血红蛋白(HbA1C),第8周时测12h尿白蛋白排泄率(UAER)、尿视黄醇结合蛋白(URBP)和UMCP-1排泄率,处死大鼠后留取肾脏标本用于逆转录聚合酶链反应半定量测定MCP-lmRNA的含量。结果:1与正常对照组(C组)比较,糖尿病组(D组)、安体舒通治疗组(S组)第8周血糖及HbAlc水平均明显升高,而两组间无显著性差异;2第8周,D、S组UAER及URBP、UMCP-1排泄率和肾脏肥大指数均高于C组,S组4项指标较D组明显降低,且UMCP-1排泄率与UAER、URBP排泄率及肾脏肥大指数呈正相关;3与C组相比,D组大鼠肾脏MCP-1mRNA表达明显上调,S组MCP-1mRNA表达也增加,但明显低于D组。结论:安体舒通对2型糖尿病大鼠肾脏有明显的保护作用,且这种保护作用是独立于降糖作用之外的。其机制可能与抑制肾组织mcp-1表达及其排泄有关。
