牙周膜牵张成骨快速移动牙牙髓中IL-8表达的变化

摘    要:目的探讨牙周膜牵张成骨后实验动物移动牙牙髓中白细胞介素8( interlukin 8, IL-8)表达的变化。方法山东本地健康杂种犬15只,随机分为5组,每组3只。4个实验组,分别为加力2周组(A组),加力后保持1周组(B组),加力后保持2周组(C组),加力后保持4周组(D组),一个对照组(E组)。拔除下颌双侧第二前磨牙,以单根的第一前磨牙作为移动牙,双根的第三前磨牙作为支抗牙,实验动物通过牙周膜牵张成骨快速移动牙齿,并分别在加力2周及加力后保持1、2、4周时结束实验,局麻下拔除移动牙,取出牙髓组织标本,采用ELISA夹心法测定移动牙牙髓中IL-8的含量及其在矫治过程中的变化。结果A组实验犬移动牙牙髓中IL-8含量与E组相比显著增高,差异具有统计学意义(P<0.05)。B组IL-8含量略有下降。C组IL-8含量显著降低,与A组比较差异具有统计学意义(P<0.05),但与E组比较差异无统计学意义(P>0.05)。结论牙周膜牵张成骨后IL-8含量升高,加力停止2周后即恢复到正常水平,提示牙周膜可以快速牵张成骨,加快牙齿移动速度。

Abstract:Objective To explore the immunohistochemical expression of IL-8 in pulpal tissues on orthodontic rapid tooth movement through distraction osteogenesis.Methods Fifteen native healthy-hybrid dogs were randomly divided into 5 groups: 3 dogs in each group.The device was activated for 2 weeks(group A),the device was fixed for 1 week(group B),the device was fixed for 2 weeks(group C),the device was fixed for 4 weeks(group D) and the control group(group E).In the experimental groups,the second premolar was extracted,and the first premolar served as the moving tooth and the third premolar served as another tooth.Then the first premolar of the experimental animals was moved by using the distraction device and then was extracted when the device had been activated for 2 weeks,fixed for 1 week,2 weeks,and 4 weeks respectively.Then immunohistochemical expression of IL-8 in pulpal tissues was determined by the ELISA method.Results The IL-8 concentrations in group A were significantly higher than that in the control group(P<0.05).After the device was fixed for 1 week,the IL-8 concentrations in group B and C,there were significant differences from group A(P<0.05)but no significant differences from the control group(P>0.05).Conclusions The immunohistochemical expression of IL-8 is significantly increased notably after experimentation,and declined to the normal level 2 weeks after the device was fixed,indicating distraction osteogenesis of the periodontal ligament can increase the orthodontic tooth movement.
