K4500型血液分析仪试剂的研究与应用

目的 :研究非氰化物溶血剂在白细胞分类计数与血红蛋白测定中的作用 ,研制适用于 K450 0血液分析仪的配套试剂。方法 :用季铵盐型表面活性剂破坏红细胞 ,进行白细胞计数与分类 ,用 SLS破坏红细胞并进行血红蛋白的测定 ,以无机盐为主体组成中性等渗的稀释液 ,建立配套的 K450 0血液分析仪检测试剂。结果 :所形成的血红蛋白衍生物 λmax=535nm,且在 5h内稳定。自制试剂的精密度试验结果表明 ,各项主要参数的相对标准偏差 CV<5%。与 sysmex试剂可比性实验表明 ,两者结果无显著性差异。结论 :采用该试剂进行血常规分析 ,方法简便实用 ,结果准确 ,效果良好

Creation and Application of K4500 Automated Heamatology Anal yzer Reagent

Objective To study the use of cyanidefree Lysing reagent on WBC diff and count and hemoglobin determination.The study suit to sysmex K4500 Automated Heamatology Analyzer.Methods Using dodecyltnmethyl ammonium chloride to make certain affection of all blood cells,and SLS to give a new product after combine with free hemoglobin andit is established a lytic system.Results The maximum peak of absorption is at 535nm and unsteady within 5 hours.Repeated examination indicated that the relative standard deviation was smaller than 5%.Comparing examination showed no significant diference between determination result of the reagent made at Xi'an create bio co.ltd.and sysmex co.ltd in Japan.Conclusion This method is characterized by using only one cyanide free reagent to count and diff WBC and determinate hemoglobin.The advantages were accurate,sensitive,convenient and suitable for automation.
