败血症小鼠肝肺组织MDA和SOD及诱导型一氧化氮合酶表达的研究

摘    要:目的探讨腹腔感染败血症小鼠肝和肺组织的病理变化、丙二醛(MDA)的含量、超氧化物歧化酶(SOD)的活性和诱导型一氧化氮合酶(iNOS)mRNA的表达及意义。方法采用盲肠结扎并穿孔(CLP)法复制败血症模型。32只小鼠随机分为对照组(假手术组)和CLP组,每组16只。模型制备成功后6h取肝、肺组织进行病理学检查,12h取肝、肺组织测定MDA含量和SOD活性,原位杂交法检测iNOS mRNA的表达。结果光镜和电镜下CLP组小鼠肝和肺组织的损伤均较对照组明显加重。CLP组肝和肺组织MDA的含量分别为(4.29±1.68)nmol/(mg.pro)和(1.93±0.13)nmol/(mg.pro),均较对照组肝组织(0.99±0.21)nmol/(mg.pro)和肺组织(1.66±0.05)nmol/(mg.pro)明显升高(P<0.05或0.01)。CLP组肝和肺组织的SOD活性分别为(201.63±45.38)u/(mg.pro)和(120.84±5.62)u/(mg.pro),均较对照组肝组织(371.62±59.98)u/(mg.pro)和肺组织(150.22±9.03)u/(mg.pro)明显降低(P<0.05或0.01)。CLP组肝和肺组织iNOSmRNA阳性表达率分别为90%和100%,均明显高于对照组(0%和10%,P<0.01)。并且,CLP组小鼠肝和肺组织iNOSmRNA的表达积分分别为(4.33±0.98)和(4.23±0.69),均明显高于对照组([0.73±0.58)和(0.97±0.88),P<0.05]。结论腹腔感染败血症小鼠肝、肺组织病理损伤严重,MDA含量升高,SOD活性降低,iNOS mRNA的表达增强。

Abstract:【Objective】 To investigate the contents of malondialdehyde(MDA),the activities of superoxide dismutase(SOD) and the expressions of inducible nitric oxide synthase(iNOS) mRNA in hepatic and lung tissues of sepsis mice.【Methods】 Thirty-two healthy female mice were randomly divided into 2 groups: ① control group received sham operation;② CLP group received a cecal ligation and puncture(CLP).Histopathological examinations of hepatic and lung were performed at 6h after operation.At 12h after operation,the contents of MDA and the activities of SOD in hepatic and lung tissues were determined.The expressions of iNOS mRNA were detected by in situ hybridization.【Results】 Compared with control group,the histopathological damages of hepatic and lung tissues of CLP group were significantly elevated.The MDA contents in hepatic tissues [(4.29±1.68) nmol/(mg·pro)] and in lung tissues [(1.93±0.13) nmol/(mg·pro)]of CLP group were significantly increased than those in control group[(0.99±0.21),(1.66±0.05) nmol/(mg·pro)](P <0.05 or 0.01).The SOD activities in hepatic tissues [(201.63±45.38) u/(mg·pro)] and in lung tissues [(120.84±5.62)u/(mg·pro)] of CLP group were decreased remarkably than those in control group[(371.62±59.98),(150.22±9.03) u/(mg·pro)](P <0.05 or 0.01).The rates of iNOS mRNA expression in hepatic tissues(90%) and in lung tissues(100%) of CLP group were significantly higher than those in control group(0%,10%)(P <0.01).Furthermore,the levels of iNOS mRNA expression in hepatic tissues(4.33±0.98) and in lung tissues(4.23±0.69) of CLP group were significantly increased than those in control group[(0.73±0.58),(0.97±0.88)](P <0.05).【Conclusion】 The contents of MDA and the expressions of iNOS mRNA increase and the SOD activities decrease in hepatic and lung tissues of sepsis mice.
