灰树花提取物对脾虚小鼠脾脏Bcl-2,Bax表达的影响

摘    要:目的:探讨灰树花提取物对脾虚小鼠脾脏凋亡相关基因Bcl-2和Bax的影响.方法:健康雄性清洁级昆明种小鼠48只随机分为6组,即空白组、脾虚组、阳性药组(香菇多糖,2 mg·kg-1)、灰树花提取物低、中、高剂量组(5,10,20 mg·kg-1),每组8只.采用番泻叶-劳倦过度复合因素法造模14 d,之后灰树花提取物和香菇多糖溶液滤菌后ip 10 d,第11天处死动物.免疫组化法观察脾脏凋亡相关蛋白Bcl-2,Bax的表达,并用逆转录聚合酶链反应(RT-PCR)测定脾细胞凋亡相关基因Bcl-2,Bax mRNA的表达情况.结果:给予灰树花提取物后,Bcl-2阳性产物平均灰度值比脾虚组明显降低(P<0.01),Bax阳性产物平均灰度值比脾虚组明显升高(P<0.01).给予灰树花提取物后,Bcl-2 mRNA表达显著升高(P<0.01),Bax mRNA表达显著降低(P<0.01).结论:灰树花提取物可以通过促进Bcl-2蛋白与mRNA的表达、抑制Bax蛋白与mRNA的表达,提高Bcl-2/Bax的比值,可以通过影响线粒体途径来抑制脾淋巴细胞凋亡的发生.

Abstract:Objective :To investigate the effect of extract of grifola frondosa on Bcl-2 and Bax in spleen of spleen deficiency mouse. Method : Forty-eight KM mice were divided into 6 groups: normal group, spleen deficiency group, lentinan group(2 mg ·kg-1), three groups of extract of grifola frondosa(5,10,20 mg ·kg-1), ip ten days. Explore the expression of Bcl-2 and Bax in spleen of spleen deficiency mice by SP immunohistochemistry staining and RT-PCR. Result : The expression of Bcl-2 decreased, and the expression of Bax increased in spleen deficiency group. After given the extract of grifola frondosa,the expression of Bcl-2 increased significantly (P<0.01), and the expression of Bax decreased significantly(P<0.01). Conclusion : The extract of grifola frondosa could maintain the immunologic function by upregulating the expression of Bcl-2 and downregulating the expression of Bax in spleen of spleen deficiency mice.
