摘    要:目的研究重组人粒细胞集落刺激因子(rhG-CSF)对HL-60细胞的影响。方法瑞氏染色观察细胞形态的变化,吖啶橙/溴化乙锭双染色观察凋亡情况,NBT和CD13测定观察成熟分化情况,MTT实验观察增殖活性。结果加入rhG-CSF 48 h后,HL-60细胞凋亡率升高,增殖活性下降,而且随着rhG-CSF浓度的升高,这种作用更加明显。成熟分化特征变化不明显。结论rhG-CSF有促进HL-60细胞凋亡并抑制其增殖的作用。

Abstract:Objective To investigate the effects of rhG-CSF on HL-60 cells. Methods HL-60 cells were cultured in different concentrations of rhG-CSF, and the cells were morphologically observed by using Wrights, acridine orange (AO) and ethidium bromide (EB) staining, MTT assay, nitroblue tetrazolium (NBT), expression of CD13. Results Apoptosis of HL-60 cells was enhanced and proliferation was inhibited after culture with rhG-CSF for 48 hours, and the enhancing and inhibiting effects were stronger with the rise of rhG-CSF concentration. Conclusion RhG-CSF could enhance the apoptosis of HL-60 cell and inhibit the proliferation.
