胰岛素可抵抗MPP^+诱导的PC12细胞的凋亡

摘    要:目的观察胰岛素在MPP+诱导PC12细胞凋亡中的干预作用.方法应用MTT法研究细胞活性的改变,应用HOECHST33258染色结合荧光显微镜技术及流式细胞技术检测不同药物对PC12细胞的凋亡诱导作用,应用半定量逆转录聚合酶链式反应(RT-PCR)测定胰岛素受体(insulin receptor,IR)mRNA的改变.结果①MPP+诱导PC12细胞凋亡,胰岛素可以抵抗此凋亡作用;②以上两种处理,均未见到胰岛素受体mRNA的改变,推测胰岛素受体的自身磷酸化有改变.结论胰岛素可以抵抗MPP+诱导的PC12细胞的凋亡.

Abstract:Aim:To study the effect of the insulin against 1-methyl-4-phenylpyridium ion (MPP +)induced apoptosis in pheochromocytoma (PC12) cells.Methods:3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) assay,HOECHST 33258 staining method,flow cytometric analysis were performed on PC 12 cells,the expression of insulin receptor (IR) mRNA was measured by using RT-PCR.Results:(1)Cell treated with MPP + exhibited various apoptotic features,while cells pretreated with insulin prior to MPP + exposure showed a decrease in the occurrence of apoptotic features.(2)Both MPP + and insulin had no effects on the expression of IRmRNA,the phosphorylation of IR may be a potential mechanism.Conclusion:The insulin may exert a protective effect against MPP +-induced apoptosis in PC12 cells.
