5-对氯苯基-1-正丁基-3-吡唑烷酮的合成及抗惊厥作用研究

摘    要:根据3-吡唑烷酮类化合物的定量构效关系,预测5-取代1-正丁基-3-吡唑烷酮类化合物的1位和5位取代基疏水性参数之和Fr约为5.6的化合物,可能具有较强的抗惊厥活性。为此,合成了具备这个条件的化合物5-对氯苯基-1-正丁基-3-吡唑烷酮,药理实验结果与预测相符,具有较强的抗惊厥活性,其ED_(50)为9.1mg/kg。

Abstract:According to the quantitative structure-activity relationship of 1, 5-substituted-3-pyrazolidinones previously synthesized by others, stronger anticonvulsant activity of the compound having ∑Fr5.6 were forecasted and such synthesized compound, 5-(4-chlorophenyl)-1-butyl3-pyrazolidinone, showed the perdicted pharmacological activity and gave ED_(50)=9.1mg/kg.
