摘    要:目的:观察绞股蓝皂苷联合银杏叶提取物对2型糖尿病(diabetes mellitus type 2,T2DM)并非酒精性脂肪性肝病(nonalcoholic fatty liver disease,NAFLD)大鼠肝组织和血浆硫化氢(H_2S)含量的影响。方法:40只SPF级雄性SD大鼠随机分为空白组,T2DM并NAFLD模型组。模型组造模周期8周。将模型大鼠随机分为模型组(予同等体积的纯净水灌胃),对照组予绞股蓝0.5 g·(kg·d)-1灌胃干预],治疗组予绞股蓝0.5 g·(kg·d)-1联合银杏叶提取物0.1 g·(kg·d)-1]灌胃干预。继续予高糖高脂饲料造模;整个实验周期共14周。检测肝组织H_2S和三酰甘油(triacylglycerol,TG)水平,检测血H_2S、血糖、TG水平。结果:1血浆H_2S浓度比较:与空白组比较,治疗组、对照组、模型组血浆H_2S下降;但治疗组、对照组高于模型组,差异有统计学意义(P0.01),治疗组高于对照组,差异有高度统计学意义(P0.01)。2肝组织H_2S含量比较:治疗组、对照组与模型组比较,差异有统计学意义(P0.01),治疗组高于对照组,差异有统计学意义(P0.01)。3空腹血糖水平比较:空白组为(5.56±0.42)mmol·L~(-1)、治疗组为(10.24±1.18)mmol·L~(-1)、对照组为(12.28±1.86)mmol·L~(-1)、模型组为(18.12±2.26)mmol·L~(-1),治疗组、对照组低于模型组,差异有统计学意义(P0.01),治疗组低于对照组,差异有统计学意义(P0.01)。4血TG水平比较:空白组(1.02±0.10)mmol·L~(-1)、治疗组(1.92±0.44)mmol·L~(-1)、对照组(2.45±0.51)mmol·L~(-1)、模型组(3.82±0.58)mmol·L~(-1),治疗组、对照组与模型组比较,差异有统计学意义(P0.01),治疗组低于对照组,差异有统计学意义(P0.01)。5肝组织TG水平:空白组(30.26±2.48)mg·g~(- 1)、模型组(228.46±8.48)mg·g~(- 1)、治疗组(153.12±9.98)mg·g~(- 1)、对照组(196.24±9.78)mg·g~(- 1),治疗组、对照组低于模型组(P0.05或P0.01),治疗组低于对照组(P0.05)。结论:绞股蓝联合银杏叶提取物能调整T2DM并NAFLD模型血糖、血脂代谢,抑制血浆和肝组织H_2S的表达。

Effect of Gypenosides and Ginkgo Biloba Extract on Liver Tissue and Plasma H2S in Type 2 Diabetes Mellitus Induced by Nonalcoholic Fatty Liver Disease in Rats

Abstract:Objective:To observe the effects of Gynostemma pentaphyllum extract combined with Ginkgo biloba extract on the liver tissue and plasma hydrogen sulfide (H2S) content in nonalcoholic fatty liver disease (NAFLD) rats with type 2 diabetes mellitus (T2DM).Methods:Forty SPF male SD rats were randomly divided into blank group,T2DM group and NAFLD model group.The model group was modeled for 8 weeks.The rats in the control group were treated with Gynostemma pentaphyllum 0.5 g · (kg ·d)-1.The rats in the treatment group were treated with Gynostemma pentaphyllum 0.5 g · (kg · d)-1 combined with Ginkgo biloba extract 0.1 g · (kg · d)-1 gavage intervention.High glucose and high fat diet were used to continue the modeling and the entire experimental cycle was of 14 weeks.The levels of H2 S,TG and TG in liver tissue were detected.Results:①Compared with the blank control group,the levels of H2S in the treatment group,the control group and the model group decreased significantly (P <0.01),but that of the treatment group and the control group were significantly higher than that of the model group (P < 0.01).②Compared with the model group,the difference of H2S content between the treatment group and the control group was statistically significant(P < 0.01),and that of the treatment group was statistically higher than that of the control group (P < 0.01).③The levels of fasting blood glucose in the control group were (5.56 ± 0.42) mmol · L-1,(10.24 ± 1.18) mmol · L-1 in the treatment group and (12.28 ± 1.86) mmol · L-1 in the control group,(18.12 ±2.26) mmol · L-1 in the treatment group,which suggested that the control group was significantly lower than the model group (P < 0.01) and the treatment group was significantly lower than the control group (P < 0.01).④Compared with the control group,the levels of TG in the control group were (1.02 ± 0.10)mmol · L-1,(1.92 ±0.44) mmol · L-1 in the treatment group and (2.45 ±0.51) mmol · L-1 in the control group 0.58 mmol ·L-1.There was significant difference between the two groups (P < 0.01).⑤The levels of TG in liver tissue were (30.26 ± 2.48) mg · g-1,(228.46 ±8.48) mg · g-1 and (153.12 ±9.98) mg · g-1,(196.24 ±9.78) mg · g-1 respectively,which meant that the treatment group and the control group were significantly lower than the model group (P < 0.05;P < 0.01) and the treatment group was significantly lower than the control group (P < 0.05).Conclusion:Gynostemma pentaphyllum extract combined with Ginkgo biloba extract can modulate the blood glucose and lipid metabolism in T2DM and NAFLD models,and inhibit the expression of H2S in plasma and liver tissue.
