君子扶正汤对胃癌大鼠细胞凋亡及血管内皮生长因子的影响

目的:探讨君子扶正汤对胃癌模型大鼠细胞凋亡及血管内皮生长因子(Vascular endothelial growth factor,VEGF)分泌的影响。方法:选取健康Wistar大鼠110只为研究对象,自由摄食、饮水,适应性饲养1周后,抽取10只生长良好的MFC胃癌小鼠的腹水,制成癌细胞悬液,接种于其余小鼠右腋皮下,建立胃癌大鼠模型。随机分为对照组和研究组,每组50只,对照组采用生理盐水灌胃,研究组采用君子扶正汤灌胃。12 d后,处死大鼠取出肿瘤组织,测定平均瘤体质量、脾指数、胸腺指数,应用酶联免疫吸附实验测定胃癌细胞上清液中VEGF的分泌情况,应用流式细胞仪检测胃癌细胞的凋亡情况。结果:研究组大鼠瘤体质量低于对照组,而脾指数、胸腺指数、SGC-7901细胞和MFC细胞凋亡率均高于对照组,差异具有统计学意义(P0.05);研究组大鼠SGC-7901细胞和MFC细胞分泌VEGF浓度分别为(158.36±20.67)ng·L~(-1)、(131.75±18.63)ng·L~(-1),均低于对照组的(205.36±23.63)ng·L~(-1)、(182.57±22.12)ng·L~(-1),差异具有统计学意义(P0.05)。结论:君子扶正汤可诱导胃癌模型大鼠细胞凋亡,抑制肿瘤细胞VEGF分泌,提高胃癌模型大鼠的免疫功能。

Effect of Junzi Fuzheng Decoction on Cell Apoptosis and Vascular Endothelial Growth Factor Secretion of Rats with Gastric Cancer

Objective To investigate the effect of Junzi Fuzheng decoction on cell apoptosis and vascular endothelial growth factor (VEGF) secretion of model rats with gastric cancer.Methods:110 healthy Wistar rats were selected as research object and they were given free feeding and drinking water.After one-week adaptive breeding,the ascites of ten well-grown MFC gastric cancer mice were extracted,and cancer cell suspension was made with saline and then inoculated into the right axilla of mouse.The model rats with gastric cancer were established and randomly divided into two groups,50 cases in each group.The control group took gavage with saline,and the research group took garage with Junzi Fuzheng decoction.After 12 d,rats were sacrificed and tumor tissues were taken out.The average tumor weight,spleen index,thymus index were measured.The VEGF secretion in supernatant of gastric cancer cell was measured by enzyme-linked immnunosorbent assay.The apoptosis of gastric cancer cell was measured by flow cytometer.Results:The tumor weight of research group was lower than that of control group,but spleen index,thymus index,SGC-7901 cell and MFC cell apoptosis rate were higher than control group (P ＜ 0.05);VEGF concentration of SGC-7901 cell and MFC cell secretion of research group were (158.36 ± 20.67) ng · L-1 and (131.75 ± 18.63) ng · L-1,which were lower than (205.36 ±23.63) ng · L-1,(182.57 ±22.12) ng· L-1 of control group (P ＜0.05).Conclusion:Junzi Fuzheng decoction can induce apoptosis of gastric cancer cell,inhibit VEGF secretion of tumor cell,increase immune function of model rats with gastric cancer.
