化瘀解毒方对胃癌大鼠血清炎症因子、免疫功能和肿瘤相关因子的影响

摘    要:目的:观察化瘀解毒方对胃癌大鼠血清炎症因子、免疫功能和肿瘤相关因子的影响,研究化瘀解毒方抗癌的作用机制。方法:48只Wistar大鼠剔除死亡及离群值后随机分为空白组、模型组、中药低剂量组、中药高剂量组,每组10只。造模成功后,中药组分别给予低、高剂量化瘀解毒方灌胃,空白组和模型组给予等剂量生理盐水灌胃;干预12周结束后取材,测定血清炎症相关细胞因子白细胞介素(interleukin-6,IL-6)、IL-10、肿瘤坏死因子(tumor necrosis factor-α,TNF-α)水平及细胞免疫、体液免疫指标,观察大鼠胃癌组织p53、STAT3、VEGF蛋白表达情况。结果:药物干预后,中药高剂量组血清中IL-6、IL-10、TNF-α、Ig G、Ig M、Ig A、CD3+、CD4+含量显著下降,CD8+水平明显升高,与模型组及中药低剂量组比较,差异有统计学意义(P0.05)。与空白组比较,模型组及各剂量中药组p53、STAT3、血管内皮生长因子(vascular endothelial growth factor,VEGF)阳性表达均显著上升(P0.05),且中药高剂量组p53、STAT3、VEGF阳性表达水平明显低于模型组及中药低剂量组,差异有统计学意义(P0.05)。结论:化瘀解毒方抗癌的作用机制与其能够提高细胞和体液免疫,减轻胃癌大鼠炎症反应,降低p53、STAT3、VEGF等肿瘤相关因子的表达有关。

Effects of Huayu Jiedu Decoction on Inflammatory Factors,Immune Function and Tumor-Related Factors in Rats with Gastric Cancer

Abstract:Objective:To observe the effect of Huayu Jiedu Decoction on serum inflammatory factors,immune function and tumor-related factors in rats with gastric cancer,and to study the anti-cancer mechanism of Huayu Jiedu Decoction.Methods:48 Wistar rats were randomly divided into blank group,model group,low dose of traditional Chinese medicine group,high dose of traditional Chinese medicine group,12 rats in each group.The rats in the untreated group were treated with low-dose and high-dose Huayu Jiedu decoction.The rats in the blank group and the model group were given normal saline by intragastric administration.After the end of 12 weeks,The levels of IL-6,IL-10,TNF-α,CD3 +,CD4 +,CDs + in peripheral blood and the levels of IgG,IgM and IgA in humoral immunity were detected by immunocytochemistry.The expression of p53,STAT3 and VEGF was detected by Western-blot.The expression of p53,STAT3 and VEGF protein were detected by HE staining and optical microscopy.Results:After the drug intervention,the serum IL-6,IL-10,TNF-α,IgG,IgM,IgA,CD3 +,CD4 + in the high dose group decreased significantly,and the level of CDs + was significantly increased,compared with the model group and the low-dose group,the difference was statistically significant (P < 0.05).Compared with the blank group,p53,STAT3,VEGF positive expression in the model group and each dose of Chinese medicine group increased significantly(P <0.05),and p53,STAT3,VEGF positive expression of high dose of Chinese medicine group was significantly lower than the model group and low dose of Chinese medicine group,the difference was statistically significant(P < 0.05).Conclusion:The antitumor mechanism of Huayu Jiedu Decoction is related to its ability to enhance cellular and humoral immunity,reduce inflammatory reaction in gastric cancer rats,and decrease the expression of p53,STAT3,VEGF and other tumor-related factors.
