摘    要:目的 比较肠出血性大肠埃希菌流行菌株的细胞毒性,筛选强毒株,为进一步研究细菌的致病机理提供理论依据.方法 采用PCR技术鉴定菌株的毒力基因E-hlyA、stx2、stxl,Giemsa染色观察细菌的黏附情况,通过测定感染后Vero细胞的乳酸脱氢酶释放情况了解细菌的Vero细胞毒性强弱.结果 肠出血性大肠埃希菌O157∶H7菌株含有的毒力基因不同,均具有较强的黏附能力和Vero细胞毒性,其中国际菌株933W/O,中国流行菌株882364、01H112和日本菌株Cua9的Vero细胞毒性较强.结论 筛选出的流行菌株强毒株,可用于进一步的细菌感染模型构建和毒力基因致病机制研究.

Abstract:Objective To compare cytotoxicity of the enterohaemorrhagic Escherichia coli (EHEC) popular strains and screen strong virulent strains, so as to provide basis for further study on the pathogenic mechanism of the bacteria. Methods Virulence genes E-hlyA, stx2, stx1 were identified by PCR technology. Bacteria adhesion was observed by Giemsa staining and lactate dehydrogenase release after Vero cells infection was detected for understanding the cytotoxicity of bacterial infection. Results EHEC popular strains contained different virulence genes, but all had strong adhesion ability and Vero cytotoxicity. The international strain 933W/O, Chinese popular strains 882364 and 01H112, and Japan strain Cua9 had stronger Vero cytotoxicity. Conclusion The strong virulent strains could be identified for further bacterial infection model construction and pathogenic mechanism research of virulence genes.
