筑巢式PCR检测白血病bcr—ablmRNA

应用筑巢式逆转录酶聚合酶链反应（ＲＴＰＣＲ）方法检测了２６例慢性粒细胞性白血病及１０例急性淋巴细胞白血病患者的ｂｃｒａｂｌｍＲＮＡ，在２１例ｐｈ阳性的慢粒和２例ｐｈ阳性的急淋患者中，检出两种ｂｃｒａｂｌ融合基因ｍＲＮＡ（１６５ｂｐ或９０ｂｐ），５例ｐｈ阴性的慢粒中，４例阳性，１例阴性，经治疗后ｐｈ染色体消失，ＲＴＰＣＲ仍能检出ｂｃｒａｂｌ融合基因。本方法具有高度敏感性，可从１０６个正常细胞ＲＮＡ中检出１个白血病细胞。是对慢粒和部分急淋的诊断及监测的一种快速、灵敏、特异的方法

Detection of bcr abl mRNA in Leukemia by RT PCR

RT PCR method was used to detect the mRNA of 26 patients with chronic myeloid leukemia (CML)and 10 patients with acute lymphocyte leukemia (ALL).The results showed that 21 philadelphia chromosome positive (ph+)patients and 2 ph+ ALL patients bcr abl fusion gene were present in all these patients.4 of 5 cytogenetically negative exhibited PCR detectabie bcr abl rearrangement. The detection of bcr abl fusion gene was rapid, sensive and specific method in the diagnosis, evaluation of treatment and monitoring of minimal residual disease.
