摘    要:应用筑巢式逆转录酶聚合酶链反应(RTPCR)方法检测了26例慢性粒细胞性白血病及10例急性淋巴细胞白血病患者的bcrablmRNA,在21例ph阳性的慢粒和2例ph阳性的急淋患者中,检出两种bcrabl融合基因mRNA(165bp或90bp),5例ph阴性的慢粒中,4例阳性,1例阴性,经治疗后ph染色体消失,RTPCR仍能检出bcrabl融合基因。本方法具有高度敏感性,可从106个正常细胞RNA中检出1个白血病细胞。是对慢粒和部分急淋的诊断及监测的一种快速、灵敏、特异的方法

Abstract:RT PCR method was used to detect the mRNA of 26 patients with chronic myeloid leukemia (CML)and 10 patients with acute lymphocyte leukemia (ALL).The results showed that 21 philadelphia chromosome positive (ph+)patients and 2 ph+ ALL patients bcr abl fusion gene were present in all these patients.4 of 5 cytogenetically negative exhibited PCR detectabie bcr abl rearrangement. The detection of bcr abl fusion gene was rapid, sensive and specific method in the diagnosis, evaluation of treatment and monitoring of minimal residual disease.
