摘    要:目的:探讨重复经颅磁刺激(repetitive transcranial magnetic stimulation,rTMS)对大鼠脑缺血后神经干细胞和新生神经细胞的影响。方法:选用成年雄性SD大鼠54只,采用开颅电凝法制作大鼠左侧局灶性大脑中动脉梗塞模型,随机分为假刺激组、高频低强度刺激组和高频高强度刺激组。在缺血后连续刺激1周和2周处死大鼠,免疫组化检测大鼠大脑缺血侧Br-du和Nestin阳性细胞表达,比较相同时间点各组间大鼠神经功能NSS评分,了解rTMS对脑缺血神经功能恢复的影响。结果:高频阈上rTMS治疗可改善脑缺血后神经功能的恢复,其治疗效应与rTMS强度和时间相关,并能有效增加缺血侧BrdU阳性细胞数和Nestin标记的新生神经细胞数(P<0.05)。结论:高频阈上rTMS可促进脑缺血损伤后内源性神经干细胞的增殖及迁移,有助于神经功能的恢复。

Abstract:Objective:To explore the repetitive transcranial magnetic stimulation(rTMS) on the impact of neural stem cells and newborn nerve cells after cerebral ischemia.Methods: Fifty-four adult male SD rats were used to make the focal middle cerebral artery occlusion model by the craniotomy electric coagulation.These were randomly divided into rTMS sham stimulation group,the high-frequency low-intensity group,and the high-frequency high-intensity group.The rats with cerebral ischemia were killed after continuous stimulation of 1w and 2w.Brdu and Nestin positive cells expression was detected by immuno-histochemical method in the side of cerebral ischemic.Neurological NSS scores in each group were contrasted to learn the impact of rTMS inducing neurological functional recovery of cerebral ischemia at the same time point.Results: Rats with cerebral ischemia were treated by high-frequency threshold stimulation.It was thought that rTMS may improve recovery of neurological function after cerebral ischemia,in which treatment effects may be associated with the stimulus intensity and time.RTMS can increase the number of BrdU-positive cells and Nestin-positive cells in the cerebral ischemic side.The number of cells increased significantly,compared with the control group(P<0.05).Conclusion: High-frequency threshold stimulation can promote the proliferation and migration of endogenous neural stem cells after cerebral ischemic injury,and is conducive to nerve function recovery.
