阿仑膦酸钠与利维爱对实验性大鼠骨质疏松症的骨矿含量和生物力学的影响

摘    要:目的:评价阿仑膦酸钠(alendronate,ALN)治疗绝经后骨质疏松症的疗效及其与利维爱(LV)的对比。方法:将Wistar雌性大鼠60 只随机分为正常对照组、去势对照(OVX)组、ALN 组、LV 组、ALN+ LV组(每组12 只)。正常对照组做腹腔开关手术,其余4 组做双侧卵巢切除手术。模型成立后,治疗6 个月,治疗过程中测量大鼠的全身和腰椎2~4 的骨密度(BMD)、股骨的极限负荷(LL)和极限挠度(LD)。结果:经治疗后,ALN组、LV组和ALN+ LV 组大鼠的全身骨密度较治疗前分别增加4.49% ,6.12% ,6.74% ,较同期OVX组分别增加13.41% ,15.85% ,15.85% ;腰椎2~4 的BMD 较治疗前分别增加4.98% ,7.73% ,7.70% ,较同期OVX 组分别增加14.29% ,16.75% ,17.24% 。ALN 组、LV 组和ALN+ LV 组大鼠股骨的极限强度和极限挠度较同期OVX组也明显恢复(P< 0.05),取得了良好的疗效。ALN 组的疗效好于LV 组。结论:ALN 是一种对治疗绝经后骨质疏松症有良好疗效的药物,其疗效优于LV,联合应用疗效更好

Abstract:Objective:Evaluating the effect of Alendoronate in the treatment of postmenopausal osteoporosis (POP) and contrasting it with Livial.Methods:Sixty female Wistar rats were randomly divided into 5 groups:control group,OVX group,LN group and ALN+LV group (12 in each group).Every group,except the control one received a sham operation,was ouarietomized bilaterally.The treatment began after the osteoporosis model was established.The bone mineral density (BMD) of body and lumbar vertebrae 2~4 was measured by dual energy X ray absoptiometry (DXA),and the limit load (LL) and limit defection (LD) of femur were measured by Biomechanical experiment machine.Results:After the treatment,the BMD of rats’ body in LV,ALN and ALN+LV group increased by 4 49%,6 12%,6 74%,respectively than before treatment, and increased by 13 41%,15 85%,15 85%,respectively than those of the corresponding OVX group.The BMD of the rats’ lumbar vertebrae 2~4 increased by 4 98%,7 73,7 70%,respectively than those before treatment,and increased by 14 29%,16 75%,17 24% respectively than those of the corresponding OVX group.The Ll and LD of the femora in LV,ALN and ALN+LV group also increased obviously than those of the corresponding OVX group ( P <0 05).The curative effect of ALN+LV group was better than that of ALN group.Conclusion:These data indicate that ALN is effective drug in therapy of POP,and the curative effect is better than that of LV.If we combined ALN and LV to use,the therapeutic would be much better.
