摘    要:目的检测磷脂爬行酶1(PLSCR1)在结直肠癌组织中的表达并探讨其与结直肠癌生物学行为的关系。方法使用Western-blot和实时定量PCR方法从蛋白和基因水平检测PLSCR1在结直肠癌组织中的表达情况,复苏冻存的PLSCR1基因和蛋白水平表达较高的和相对较低的结直肠癌组织行原代细胞培养,使用细胞侵袭实验方法检测PLSCR1表达差异的结直肠癌原代细胞的侵袭行为。结果 Western-blot检测结果显示:PLSCR1蛋白在结直肠癌原发灶(R=0.536±0.056)中的表达水平显著高于正常结直肠黏膜(R=0.262±0.096)(P<0.01)。实时定量PCR检测显示:PLSCR1mRNA的相对含量在结直肠癌原发灶(RQ=4.45±0.30)中的水平显著高于正常结直肠黏膜标本(RQ=1.39±0.58)(P<0.01)。细胞侵袭实验检测发现,PLSCR1表达较高的结直肠癌原代细胞穿膜细胞数,显著高于PLSCR1表达相对较低的结直肠癌原代细胞(P<0.01)。结论 PLSCR1的异常表达在结直肠癌的发生和侵袭过程中发挥着重要作用。

Increased expression of PLSCR1 in colorectal cancer and its effects on invasion of cancer cells

Abstract:Objective To detect the expression of phospholipid scramblase 1(PLSCR1) in colorectal cancer (CRC) tissues and investigate its effects on the invasion of cancer cells. Methods Western blotting was used to detect the expression of PLSCR1 in both CRC tissues and corresponded normal colorectal mucosa. Real-time quantitative PCR was used to detect the mRNA of PLSCR1 in CRC tissues and normal colorectal mucosa. Primary culture of CRC cells was carried out in vitro, and Transwell assay was used to detect the invasion ability of CRC cells with different expression levels of PLSCR1. Results The expression level of PLSCR1 protein and the mRNA level of PLSCR1 were significantly increased in CRC tissues compared with normal colorectal mucosa(P<0.01). In the invasion test, the number of invaded cells in group of high level of PLSCR1 was significantly higher than that in group of relative low level of PLSCR1(P<0.01). Conclusion The increased expression of PLSCR1 might play an important role in genesis and invasion of CRC.
