胃淋巴瘤Survivin表达与细胞凋亡及幽门螺杆菌感染相关性研究

目的检测凋亡抑制基因Survivin在胃淋巴瘤组织中的表达,观察其与细胞凋亡及幽门螺杆菌(Helicobacterpylori,HP)感染的相关性,探讨Survivin基因在胃淋巴瘤发病过程中的作用。方法应用TdT介导的dUTP缺口末端标记(TUNNEL)技术和免疫组织化学(Elivision)法观察30例正常胃粘膜组织,30例胃淋巴瘤组织中细胞凋亡和Survivin基因及HP感染阳性表达。结果胃淋巴瘤中Survivin基因表达阳性率为46.7%(14/30),而正常胃组织均无表达,两组比较有显著性差异(χ2=18.26,P<0.01)。Survivin表达与淋巴瘤分化程度、淋巴结转移及预后相关。胃淋巴瘤组织中HP阳性检出率为36.7%(11/30),正常胃组织检出率为23.3%(7/30),两者无显著性差异(χ2=1.27,P>0.05)。胃MALT型淋巴瘤Survivin阳性表达与HP感染有显著相关性(r=0.644,P<0.05)。胃淋巴瘤组织中细胞凋亡指数(apoptosisindex,AI)显著低于正常胃粘膜,胃淋巴瘤Survivin阴性表达组细胞凋亡指数与Survivin阳性表达组比较,差异有显著性(P<0.01)。结论Survivin基因在胃淋巴瘤中表达上调,而且与分化程度、淋巴结转移及预后有关。胃淋巴瘤与Survivin表达、细胞凋亡、HP感染有关。

Study on the Expression of Survivin and it's Relativity with Apoptosis and Helicobacter Pylori Infection in Gastric lymphoma

Objective To investigate expression of survivin and it`s relation with apoptosis and infection of helicobacter pylori (HP) in gastric lymphoma.Methods Terminal deoxynudeotidyl transferase-mediated dUTP nick end labeling(TUNNEL) and immunohistochemistry Elivision method were used to determine expression of survivin,apoptotic cell and HP-infection positive-expression in 30 cases of gastric lymphoma and 30 cases of normal gastric mucosa.HP was detected by histological fuchsine staining.Results Survivin were expressed in 14 of 30 cases gastric lymphoma(46.67%) and did not found in any of 30 cases of normal control(P<0.01).HP was detected in 11 of 30 cases (36.67%) and 6 cases (30%)in the controls.The expression of survivin in gastric MALT lymphoma was related to HP infection.The apoptosis index (AI) of contrast group was higher than that in gastric lymphoma group.To gastric lymphoma,the positive-expression of survivin were significantly lower than the negative-expression of survivin for AI (P<0.01).Conclusion The increasing expression of survivin gene in gastric lymphoma suggested that survivin gene played an important role in the development of gastic lymphoma.Survivin gene expression was associated with cell apoptosis and HP infection in gastric lymphoma. 【
