内皮素、降钙素基因相关肽在肾性高血压大鼠的动态变化

摘    要:目的观察2、4、6周肾性高血压(RH)大鼠血浆及主动脉组织中内皮素(endothelin,ET)及降钙素基因相关肽(calcitoningene-relatedpeptide,CGRP)的动态变化,探讨其在RH发病中的作用。方法用颈动脉直接插管法测定大鼠血压的变化。用放射免疫测定技术测定ET、CGRP水平。结果RH组大鼠术后2周血压即已升高,4、6周时显著升高。各时期RH组大鼠血浆中CGRP含量明显低于同期对照组,而主动脉组织中CGRP含量显著高于同期对照组;血浆及主动脉组织中ET含量均显著高于同期对照组。结论RH时,血管内皮细胞功能紊乱,缩血管物质ET生成异常增多;组织中CGRP的释放减少和循环血中其含量不足均与RH发病有关。

Abstract:Objective To observe the dynamic change of endothelin(ET) and calcitonin generelated peptide(CGRP) in plasma and aortic tissue in renal hypertensive rats in order to explore the roles of them in the initiation and maintenance of renal hypertension. Methods Blood pressure,ET and CGRP were measured by inserting a cannula into the carotid artery and radioimmunoassay respectively. Results The blood pressure increased in 2 weeks and markedly increased in 4,6 weeks in RH rats. The plasma level of CGRP was significantly decreased but the level of CGRP in aorta tissue was markedly higher in 2,4,6 weeks RH rats than that of control group. Meanwhile,the contents of ET in plasma and aortic tissue were markedly increased in RH group.Conclusions The vascular endothelial cells function of renal hypertension rats was disturbed. The high levels of ET and the decreased release of CGRP from tissues and low plasma levels of CGRP might be related to the pathogenesis of renal hypertension.
