碘[125I]粒子植入对人肝癌细胞裸鼠HepG2移植瘤抑制作用的实验研究

摘    要:目的]探讨不同剂量碘125I]粒子组织间植入对人肝癌细胞裸鼠HepG2移植瘤生长的抑制作用。方法]构建20只人肝癌HepG2的裸鼠移植瘤模型,随机分为4组,每组5只。3个治疗组分别接受18.5MBq(A组)、29.6MBq(B组)、37MBq(C组)的碘125I]粒子治疗,对照组(D组)不接受治疗。采用经皮肿瘤组织直接植入的方式分别将不同剂量的碘125I]粒子植入各治疗组小鼠的瘤体内,每只小鼠瘤体内植入一粒碘125I]粒子。观察各组小鼠的体重及瘤体体积的变化。至治疗第28d时处死所有小鼠,剥离肿瘤并称重。结果]各治疗组小鼠瘤体的体积增长速度随碘125I]粒子的剂量增加而逐渐降低。A、B、C组小鼠与D组小鼠的肿瘤体积相比,抑瘤率分别是41.81%、64.45%和69.69%;A、B、C组小鼠与D组小鼠的肿瘤重量相比,抑瘤率分别是31.11%、62.22%和64.44%。结论]碘125I]粒子组织间植入治疗可抑制荷HepG2肝癌裸鼠肿瘤的生长,抑瘤作用与剂量呈正相关。

Inhibitory Effect of 125I Seed Interstitial Brachytherapy on Human Hepatocarcinoma Cell HepG2 Transplanted Tumor in Nude Mice

Abstract:Purpose] To investigate the effect of 125I seed interstitial brachytherapy with different doses on human HepG2 cells in nude mice transplanted tumor.Methods] Twenty nude mice bearing human hepatoma HepG2 cells were randomly divided into 4 groups (each n=5) and given 125I seed interstitial brachytherapy with doses including 18.5MBq 125I (group A),29.6MBq 125I (group B),37MBq125I (group C) and control (group D).125I seeds were percutaneously implanted into tumors,each nude mouse with one 125I seed implanted.The weight of each nude mouse and tumor volume were observed.At the 28th day of treatment,the mice were executed,tumor were removed and weighted.Results] Tumor growth rate decreased with increase 125I seed doses.The inhibitory rates of tumor volume in group A,B and C were 41.81%,64.45% and 69.69%,respectively.The inhibitory rates of tumor weight in group A,B and C were 31.11%,62.22% and 64.44%,respectively.Conclusion] 125I seed implantation inhibits tumor growth in HepG2 hepatoma bearing nude mice with a dose-dependent manner.
