摘    要:目的:了解Hedgehog(Hh)-Gli信号通路失调控在肝细胞癌(HCC)发生、发展中的作用及其分子靶向治疗的研究现状。方法:以Hedge-hog、肝癌/hepatocellular carcinoma(HCC)为关键词,使用CNKI(1994-2009)、NCBI(1974-2009)生物信息学数据库对已发表的文章进行检索,共检索到英文27篇,中文7篇,排除重复研究和与本研究无关的文献,最终纳入分析27篇文章。结果:研究发现,Hh信号通路不仅调控胚胎时期各胚层组织发育,而且与肿瘤发生、发展密切相关。Gli作为脊椎动物Hh信号通路锌指核转录因子,与Hh通路下游基因的特定序列结合,直接调控Hh信号通路目的基因的转录表达,在Hh信号通路调控中起到枢纽作用。Hh-Gli信号转导通路的异常激活参与HCC的发生及恶性生物学特性的维持。环杷明和(或)Gli-si RNA可以阻断Hh信号通路抑制HCC生长、侵袭和运动潜能。结论:Hh通路的激活是HCC侵袭、发展的重要因素,阻断Hh通路可能是一种新的潜在的HCC靶向治疗策略,具有临床应用价值。
