青藤碱对类风湿关节炎患者滑膜细胞凋亡细胞周期及bal-2蛋白的影响

摘    要:目的探讨中药青藤碱(SN)诱导类风湿关节炎(RA)患者滑膜细胞凋亡的发生机制,为RA治疗提供新的理论基础。方法光镜和荧光显微镜观察滑膜细胞凋亡形态的变化,流式细胞仪检测细胞凋亡率、细胞周期及bcl-2基因的表达。结果光镜及荧光显微镜下可见核固缩,变圆,荧光染色增强等典型凋亡形态学变化。流式细胞仪结果显示,在2.8~3.2mmol/L浓度范围内,随着SN浓度的增强,滑膜细胞的凋亡率增加。作用48h后,bcl-2蛋白在凋亡过程中的表达持续下调,细胞周期阻滞在G1~S期。结论SN在一定浓度及作用时间内可诱导RA患者滑膜细胞凋亡,其作用机制可能与下调bcl-2蛋白表达及阻滞G1期细胞向S期移行有关。

Abstract:Objective To explore the mechanism of sinomenine(SN) on synoviocyte apoptosis in patients with rheumatoid arthritis(RA) and provide new evidence for treatment of this condition.Methods The morphological changes of synoviocytes were observed by inverted microscope and fluorescence microscope.Synoviocyte apoptosis,changes of cell cycle and apoptosis-associated protein bcl-2 expression were measured by flow cytometry.Results The morphological changes typical of apoptosis,such as cell rounding,karyopyknosis and fluorescence enhancement,were observed.Flow cytometry showed,within a range of SN level from 2.8 mmol/L to 3.2 mmol/L,cell apoptosis rate increased in a concentration-dependent manner.The expression of Bcl-2 protein was down-regulated and the cell cycle blocked on G1 to S phase by SN lasting 48 h.Conclusion Given proper concentration and time of treatment,SN appears to induce synoviocytes apoptosis,which may be related with inhibited cell cycle from G1 to S and decreased expression of bcl-2 protein.
