肝素酶和碱性成纤维细胞生长因子与胃癌转移和预后的关系

摘    要:目的探讨肝素酶和碱性成纤维细胞生长因子(bFGF)在胃癌组织中表达及其与大肠癌临床病理特征之间的关系。方法采用免疫组织化学方法检测80例胃癌石蜡切片和20例癌旁正常胃黏膜中肝素酶和bFGF的表达情况,分析两者分别表达及共表达与胃癌患者年龄、性别、分期、分化程度、浸润深度、淋巴结转移的关系。结果免疫组织化学染色结果表明:80例胃癌中有49例(61%)呈肝素酶阳性表达,52例(65%)呈bFGF阳性表达,主要表达在癌细胞质和(或)细胞膜中,在正常胃黏膜上皮则呈阴性表达,肝素酶和bFGF在胃癌中表达明显增高(P<0.05),且两者的表达具有明显的相关性(P<0.05)。两者单独表达和共表达均与胃癌的分化程度、分期、浸润深度、淋巴结转移有关,与患者年龄、性别无关。结论肝素酶和bFGF蛋白在胃癌组织中的表达较正常胃黏膜上皮明显上调,两者单独表达和共表达与胃癌的侵袭及转移能力密切相关。

Abstract:Objective To assess protein expression levels of heparanase and basic fibroblast growth factor(bFGF)and the relation between their expression and biological factors in human gastric carcinoma.Methods A total of 80 paraffin-embedded gastric cancer and 20 normal specimens were studied by immunohistochemisty to evaluate the protein expression status of heparanase and bFGF.The relation was analyzed among the protein expression and age,sex,TNM stage,cell differentiation,invasion depth,lymphatic metastasis.Results Immunohistochemistry showed that 49 specimens were heparanase positive and 52 ones were bFGF positive among 80 cancer specimens.Both human heparanase and bFGF were highly expressed in cancer cell in contrast to no expression in normal tissue.Expression was in both cytoplasm and membranes.A significant correlation was found between the expression of either protein and cell differentiation,TNM stage,invasion depth and lymphatic metastasis.Conclusion Both heparanase and bFGF were highly expressed in gastric cancer.They may play important roles in tumor invasion,metastasis and angiogenesis.
