摘    要:目的 探讨RT-PCR法分析肿瘤细胞人端粒酶逆转录酶(hTERT)-mRNA表达水平的临床应用价值。方法 以β-actin为内对照,采用RT-PCR法测定K562、HL-60细胞及正常人外周血的hTERT-mRNA表达水平。结果 K562、HL-60细胞hTERT表现出高的表达水平,相反,正常人外周血无或低表达。结论 RT-PCR分析hTERT-mRNA表达的方法具有特异性、敏感性高,易定量,操作简单,快速等特点,可应用于临床。

Abstract:Objective To study the clinical application of RT PCR to analysis of the expression levels of human telomerase catalytic subunit(hTERT) mRNA.Methods We examined the expression levels of hTERT in K562, HL 60 cells and peripheral blood (PB) specimens from 11 normal children using semi quantitative RT PCR.Results The expression of hTERT was at significantly higher levels in K562, HL 60 cells compared with PB specimens of normal children (P<0.001). hTERT expression was nearly undetectable in PB specimens.Conclusion The results suggest that RT PCR method is an ideal for detecting the expression of hTERT in leukemia cells. It is specific, sensitive, quantitative, simple and quick, and can be nsed in clinical implications.
