益气活血方对脑缺血大鼠脑组织Glu、GABA及NR1表达的影响

摘    要:目的观察益气活血方对急性脑缺血再灌注大鼠缺血脑组织谷氨酸(glutamate,Glu)、γ-氨基丁酸(γ-aminobutyrate acid,GABA)含量及神经受体1(neuroreceptor1,NR1)表达的影响。方法采用线栓法阻断大鼠左侧大脑中动脉复制局灶性脑缺血再灌注模型。大鼠随机分为假手术组、模型组、益气活血汤组。采用高效液相色谱法和链霉亲合素-生物素酶复合物(streptavidin-biotinidase complex,SABC)染色法,分别检测Glu、GABA含量和NR1表达水平。结果脑缺血2h再灌注各时间点与假手术组比较,模型组Glu、GABA含量和NR1表达均显著升高(P〈0.05,P〈0.01);与模型组比较,治疗组脑缺血后6,48,72hGlu、NR1水平显著降低;72h GABA含量显著提高(P〈0.01)。结论益气活血方可能通过调节兴奋性氨基酸Glu和抑制性氨基酸GABA含量的平衡,及减少NR1表达防止脑缺血再灌注损伤。

Abstract:Objective To observe the effects of Qi-invigorating and blood circulation-activating(QIBCA) Recipes on the contents of glutamate(Glu),γ-aminobutyrate acid(GABA) and the expression of neuroreceptor 1(NR1) in cerebral tissue of rats after focal cerebral ischemia and reperfusion(I-R).Methods The focal cerebral I-R model was induced by ligation of the middle cerebral arteries(MCA).The animals were divided into the sham operation group,the model group,the QIBCA Recipes group.The contents of Glu and GABA were assayed by high performance liquid chromatography,and the expression of NR1 was observed using streptavidin-biotinidase complex(SABC) method.Results Compared with those of the sham operation group,the contents of Glu,GABA and the expression of NR1 were increased significantly at all intervals after I-R injury in model group(P<0.05,or P<0.01).Compared with those of the model group,the contents of Glu and the expression of NR1 were significantly decreased at all intervals after I-R injury in the treatment group.The content of GABA was increased significantly 72 hours after reperfusion in the treatment group(P<0.01).Conclusion The QIBCA Recipe could protect the cerebral tissue from I-R injury by regulating the content balance of Glu,an excitatory amino acid,and GABA,an inhibitory amino acid,and reducing the expression of NR1.
