摘    要:目的:观察加味小陷胸汤含药血清对牛主动脉内皮细胞(BAECs)的保护作用。方法:以原代培养的牛主动脉内皮细胞为研究对象,采用MTT法检测细胞生长活性,WesternBlot法检测内皮型一氧化氮合酶(eNOS)的表达。结果:一定浓度(5%、10%、15%)的加味小陷胸汤含药血清能明显促进牛主动脉内皮细胞增殖和eNOS蛋白表达,高浓度(20%)的加味小陷胸汤含药血清能抑制细胞增殖和eNOS蛋白表达。结论:加味小陷胸汤可通过促进牛主动脉内皮细胞增殖和eNOS的表达而起到保护血管内皮细胞的作用。

Abstract:Objective:To observe the protective effect of drug serum of Jiawei Xiaoxianxiong Decoction(JXD) on Bovine Aortic Endothelial Cells(BAECs). Methods:BAECs were isolated and cultured, and all experiments were performed on confluent monolayer at passengers 3-5. The cells were incubated with drug serum of JXD at different concentrations for 10 hours, the effects of drug serum of JXD on eNOS and cell proliferation were determined by Western Blot and MTT assays. Results:Drug serums of 5%, 10%, 15% JXD upregulated significantly eNOS expression and BAECs proliferation, but serum of 20% JXD had inhibition effect. Conclusions:JXD can improve BAECs proliferation and upregulates eNOS expression, so it has protective effect on BAECs.
