翻白草总黄酮降血糖作用的药效学研究

目的:研究翻白草总黄酮富集物降血糖药理作用。方法:以小剂量链脲佐菌素(STZ)腹腔注射加高脂饲料诱导2型糖尿病大鼠模型,灌胃给予高剂量36g.(kg·d)-1]、中剂量18g.(kg·d)-1]和低剂量9g.(kg·d)-1]的翻白草总黄酮富集物,测定大鼠的空腹血糖(FBG)、糖耐量2h血糖、血清胰岛素水平(FINS)、胰岛素敏感性指数(ISI)、超氧化物歧化酶(SOD)、丙二醛(MDA),并用HE染色法对大鼠进行病理切片观察。结果:中、高剂量的翻白草总黄酮能显著降低糖尿病大鼠的空腹血糖值和血清胰岛素水平,改善糖耐量,提高胰岛素敏感指数,减轻胰岛素抵抗,提高机体SOD活性,降低血清MDA含量,并对胰岛细胞起到保护修复作用。结论:翻白草总黄酮通过减轻胰岛素抵抗和保护修复胰岛细胞多种机制治疗2型糖尿病。

Study on Pharmacology of Decreasing Blood Glucose of Flavonoid from Potentilla Discolor Bunge

Objective：To study the pharmacology of Decreasing Blood Glucose effects of the total flavonoids from Potentilla Discolor Bunge. Methods：The type 2 diabetes models in rats is made by fedding high lipids foods together with injecting the streptozotocin（STZ） of small dose,then perfusing the rat＇s stomach with the total flavonoids of Potentilla discolor Bung in high doze36g.（kg·d） -1]、medium doze18g.（kg·d） -1]and low doze9g.（kg·d） -1],determinating the fasted blood glucose（FBG）,the glucose tolerance test（2HPG）,the fasting insulin levels（FINS）,the insulin sensitivity index （ISI）,the activity of SOD,and the amount of MDA,and detecting the pancreas islet B cells by HE. Results：the total flavonoids of Potentilla discolor Bung in high doze and medium doze had significantly effect of decreasing FBG and FINS on diabetic rats,improving glucose tolerance and insulin sensitivity index and reducing insulin resistance. Potentilla discolor Bung can increase the activity of SOD,decrease the amount of MDA,protect and repair pancreas islet cells. Conclusion：the total flavonoids of Potentilla discolor Bung can treat the two type diabetes mellitus by decreasing the insulin resistance ,protecting and repairing the islet cell.
