摘    要:目的检测尖锐湿疣(condyloma acuminatum,CA)患者外周血微小RNA-155(miR-155)的表达水平,并分析其与辅助性T细胞(Th)17淋巴细胞的相关性.方法选取2015年3月~2016年7月内蒙古医科大学附属医院确诊收治的CA患者50例,选取同期健康体检者50例作为对照组,实时定量PCR(qRT-PCR)法检测单一核细胞(peripheral blood mononuclear cell,PBML)中miR-155、RORγt、IL-17 mR-NA表达水平,采用流式细胞术检测外周血Th17细胞比例,采用酶联免疫吸附法(ELSA)检测血清IL-17水平.结果CA患者PBMC中miR-155、RORγt、IL-17 mRNA表达水平、Th17细胞比例显著高于健康对照组(P<0.05);CA患者血清IL-17水平显著高于健康对照组(P<0.05);Pearson检验结果显示,CA患者外周血miR-155与RORγt、IL-17 mRNA表达水平、Th细胞比例、血清IL-17水平均正相关(P<0.05).结论CA患者外周血中miR-155表达水平显著升高,与Th17细胞比列关系密切,可能与CA发病有关.

Abstract:Objective To detect the expression of micro RNA-155 ( miR-155) in peripheral blood of patients with condyloma acuminatum ( CA) and analyze its correlation with T helper cell ( Th) 17 lymphocyte.Methods Fifty cases of CA admitted to Affiliated Hospital of Inner Mongolia Medical University from March 2015 to July 2016 were selected, and 50 healthy persons in the same period were selected as control group.Real-time quantitative PCR ( qRT-PCR) was used to detect the expression levels of miR-155, RORγT, and IL-17 mRNA in mononuclear cells ( PBMC), the proportion of Th17 cells in peripheral blood was detected by flow cytometry.enzyme-linked immu-nosorbent assay ( ELSA) was used to detect serum IL-17 level.Results The expression levels of miR-155, RORγt, IL-17 mRNA, and the proportion of Th17 cells in peripheral blood mononucle-ar cell ( PBMC) were significantly higher in CA patients than in healthy controls (P<0.05) . Se-rum IL-17 level was significantly higher in CA patients than in healthy controls ( P<0.05) . Pear-son test showed that there were positive correlations between miR-155 and RORγt, IL-17 mRNA ex-pression, the proportion of Th17 cells, serum IL-17 level in CA patients (P<0.05) . Conclusion The expression level of miR-155 in peripheral blood of patients with CA is increased significantly, and it is closely related to proportion of Th17 cells, which may be related to the pathogenesis of CA.
