天钩降压胶囊中钩藤碱、异钩藤碱大鼠 在体肠吸收动力学研究

摘    要:目的:研究天钩降压胶囊中钩藤碱、异钩藤碱在大鼠肠道的吸收动力学。方法:采用大鼠在体肠灌流试验,利用HPLC测定钩藤碱、异钩藤碱的含量KROMASIL C18色谱柱(4.6 mm×200 mm,5μm),流动相乙腈-水(含0.01 mol.L-1三乙胺,冰醋酸调pH 7.5)(45∶55),流速1 mL.min-1,检测波长254 nm,柱温30℃,进样量20μL,用紫外-可见分光光度法在560nm测定酚红的含量。结果:钩藤碱吸收动力学方程为Y=-0.331 7X-0.771 8(r=0.998 5),Ka=0.331 7(h-1),t1/2=2.09(h);异钩藤碱吸收动力学方程为Y=-0.687 0X-0.150 4(r=0.985 2),Ka=0.687 0(h-1),t1/2=1.01(h)。结论:天钩降压胶囊中钩藤碱、异钩藤碱在大鼠在体小肠的吸收呈一级动力学过程。

Abstract:Objective: To study in vivo intestinal absorption kinetics of rhynchophylline and isorhynchophylline of Tiangou Jiangya capsules. Method: HPLC method was used to determine the content of rhynchophylline, isorhynchophylline and Ultra-violet (UV) spectrophotometry was used to determine the content of phenol red in the rat in vivo intestinal perfusion tests. Result: The regression of rhynchophylline is Y=-0.331 7X-0.771 8(r=0.998 5), Ka=0.331 7(h-1), t1/2=2.09(h). The regression of isorhynchophylline is Y=-0.687 0X-0.150 4(r=0.985 2), Ka=0.687 0(h-1), t1/2=1.01(h). Conclusion: The absorption of rhynchophylline and isorhynchophylline of Tiangou Jiangya capsules in rat's intestine comply with the first-order kinetics process.
