益气解毒方对大鼠局灶性脑缺血氧化应激的影响

摘    要:目的:研究中药益气解毒方对大鼠局灶性脑缺血后氧化应激损伤的影响。方法:取健康雄性SD大鼠60只,随机分为6组:假手术组、模型组、阳性药银杏叶提取物组(4 mg.kg-1)、益气解毒高、中、低剂量组(25,5,1 mg.kg-1)。给药组于手术麻醉前10 min灌胃给药,用线栓法制作大鼠大脑中动脉闭塞(middle cerebral artery occlusion,MCAO)模型,12 h后取血清和缺血脑组织制备组织匀浆液,用酶标仪定量检测超氧化物歧化酶(SOD)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、单胺氧化酶(MAO)、髓过氧化物酶(MPO)。结果:与假手术组比较,模型组脑组织SOD和GSH-Px活性明显降低(P<0.01),MDA含量和MAO,MPO活性明显升高(P<0.05)。与模型组比较,益气解毒方高、中、低剂量组、阳性药组SOD和GSH-Px显著性升高(P<0.01),MAO,MDA和MPO显著性降低(P<0.05,P<0.01)。结论:益气解毒方对大鼠局灶性脑缺血有保护作用,其作用机制可能与抗氧化应激作用有关。

Abstract:Objective: To study effect of Yiqi Jiedu formula on oxidative stress in rat focal cerebral ischemia injury model. Method: SD rats were randomly divided into Yiqi Jiedu groups(25, 5, 1 mg·kg-1), the model group, sham operation group, and positive control group(4 mg·kg-1).Rsts were given drugs before anesthesia and establish middle cerebral artery occlusion (MCAO) model.The microplate reader was used to detect superoxide dismutase(SOD), malondialdehyde(MDA), glutathione peroxidase(GSH-Px), monoamine oxidase(MAO) and myeloperoxidase(MPO) in the serum and cerebral homogenate. Result: Compared with the sham group, in model group SOD and GSH-Px decreased(P<0.01), the content of MDA and the activity of MAO and MPO increased(P<0.05).Compared with the model group, SOD and GSH-Px of Yiqi Jiedu groups and positive group were significantly higher(P<0.01), however MAO, MDA and MPO were decreased (P<0.05, P<0.01). Conclusion: Yiqi Jiedu formula can exert protection antion in focal cerebral ischemia rats, the mechanism may be related to raising efficiency and reducing toxicity.
