摘    要:目的探讨人重组骨形成蛋白一2 (rhBMP-2)对NU13T3成纤维细胞生物学行为的影响。方法培养 NIH3T3细胞,加人外源性rhBMP-2,用生长曲线、唾哇蓝(MIT)比色、流式细胞仪(FCM)分析、BrdU检测观察细胞的增殖和DNA合成,并检测碱性磷酸酶(ALP)活性和骨钙素(OC)含量观察细胞是否有成骨趋向。结果加药组细胞的增殖活性下降、DNA合成变慢,All]活性和OC含量均增高。结论rhBMP-2使NIH3T3细胞增殖活性变弱,表现成骨细胞表型。

Abstract:OBJECTIVE: To examine the effect of recombinant human bone morphogenetic protein-2 (rhBMP-2) on NIH3T3 cells. METHODS: The NIH3T3 cells were cultured in DMEM with 50 microg/L rhBMP-2. The proliferation ability, alkaline phosphatase (ALP) activity and osteocalcin (OC) quantity of NIH3T3 cells were investigated. RESULTS: The cells showed decreased proliferation ability but enhanced ALP activity and OC quantity. CONCLUSION: 50 microg/L rhBMP-2 can transform NIH3T3 fibroblasts to osteo-like cells.
