SIRT1过表达是进展期结直肠癌的一个不良预后因素

背景 SIRT1在各种生理过程包括癌症发挥重要的作用，但是在结直肠癌中的作用尚不明确。方法 采用免疫组织化学方法检测120对结直肠癌肿瘤组织和正常粘膜的组织芯片中SIRT1蛋白的表达情况。然后分析SIRT1表达水平与临床病理参数和预后的相关性。结果 所有标本SIRT1表达阳性，而且全部位于胞浆而不是核内。癌组织和正常组织之间SIRT1表达水平有明显差异。SIRT1在进展期肿瘤中过表达更明显。SIRT1过表达者的总生存期OS和无病生存期DFS明显差于低表达者。结论 SIRT1过表达与进展期结直肠癌和不良预后相关，SIRT1可能在结直肠癌进展中发挥着重要作用。

Background SIRT1 has been reported to have diverse roles in various biological processes including cancer. However, the role of SIRT1 in colorectal cancer remains unclear. Methods SIRT1 protein expression in a tissue microarray, including 120 paired colorectal cancer and normal mucosa tissues, was measured by immunohistochemistry. The correlations among SIRT1 expression, clinicopathological features, and prognosis were analyzed. Results All samples were positive for SIRT1 in the cytoplasm rather than the nucleus. There was significant difference in SIRT1 overexpression between cancer and normal tissue (P<0.01). SIRT1 overexpression was more frequently observed in advanced-stage tumors (P=0.046, 0.002). SIRT1 overexpression was significantly correlated with poor overall survival (P=0.013) and disease-free survival (P=0.012) Conclusions SIRT1 overexpression was correlated with advanced stage and poor prognosis. SIRT1 may play an import role in the progression of colorectal cancer.
