摘    要:目的:观察转染含人癌胚抗原(CEA)真核表达质粒pcDNA3-hCEA的人树突状细胞(DC)对胃癌细胞MGC803的抑制作用.方法:采用细胞因子rhIL-4和rhGM-CSF诱导培养法制备人外周血DC并用Lipofectamine向人DC转染pcDNA3-hCEA.RT-PCR检测转染细胞hCEA的表达,观察DC-hCEA细胞诱导的致敏的T淋巴细胞(CTL)对胃癌细胞MGC803的杀伤效应.结果:DC-hCEA诱导的CTL对胃癌细胞MGC803的杀伤效应强于DC(P<0.05).结论:编码CEA基因的真核表达质粒转染的人DC能够诱导较强的特异性抗肿瘤免疫效应.

Abstract:Aim: To observe the dendritic cell (DC) transfected with pcDNA3-hCEA and its immunity effect on MGC803. Methods: DC was generated from human fresh peripheral blood induced by rhGM-CSF and rhIL-4. DC vaccine was prepared by transfection with pcDNA3-hCEA using lipofection. The immunity effect of the DC on MGC803 was observed.Results: Compared with DC that was not transfectd,the antitumor immune response induced by DC-hCEA was more powerful(P<0.05). Conclusion: The results demonstrate that specific antitumor immune responses could be induced efficiently by vaccination of DC transfected with pcDNA3-hCEA.
