摘    要:目的:通过检测过氧化物酶体增生物激活受体(PPARγ)在人不同肺组织中的表达,探讨PPARγ在肺癌及肺炎性病变中的意义。方法:采用免疫组化法检测29例肺癌、7例肺良性肿瘤、15例肺炎性组织及7例正常肺组织中的PPARγ表达,并通过彩色医学图文分析系统检测PPARγ平均吸光度值。结果:PPARγ在肺癌及非肺癌组织中均有表达,正常肺组织主要表达于细胞核。细胞核中的PPARγ表达水平在正常肺组织显著高于肺癌及肺炎性组织,在肺良性肿瘤显著高于肺癌组织,差异均有统计学意义(P〈0.05)。肺癌组织中PPARγ表达从高到低依次为鳞癌、腺癌、大细胞癌、小细胞癌,鳞癌和腺癌显著高于大细胞癌和小细胞癌,中分化肺癌显著高于低分化肺癌。结论:PPARγ表达与肺癌及肺组织炎症反应有关。PPARγ可能参与了肺癌及肺组织炎症反应的发病。PPARγ表达高低反应了肺癌分化程度,其值越低恶性程度越高,可作为判断肺癌预后的指标之一。

Abstract:Objective: To study the significance of PPART in lung cancer and lung inflammatory tissues through detecting the expression of peroxisome proliferator activated receptor gamma (PPARγ) in human lung cancer and other different lung tissues. Methods: Expressions of PPARy in 29 cases of lung cancer tissues,7 cases of lung carcinoid, 15 lung inflammatory tissues and 7 cases of normal lung tissues were detected using immunohistochemistry. Average OD values were detected using chromatic medical image analysis. Results: PPARy was observed in all lung cancer tissues and non-lung cancer tissues. In normal lung tissues, PPARy was mainly expressed in nucleus. In nucleus, the PPARγ level of normal lung tissues was much higher than that of lung cancer tissues and lung inflammatory tissues, the PPARy level of lung carcinoid tissues was much higher than that of lung cancer tissues. All the differences are of statistical significance. The sequence of PPARy expression from high to low levels were squamous carcinoma, adenocarcinoma, large-cell lung cancer and small-cell lung cancer in turns. The PPARy expresion of squamous carcinoma and adenocarcinoma were significantly higher than those of large-cell lung cancer and smallcell lung cancer. The PPARγ expression in moderatedly differentiated lung cancer tissues were significantly higher than that in poorly differentiated ones. Conclusion: PPARy expression is associated with lung cancer and lung inflammatory effects, and PPARy may participate the inflammatory effects in lung tissues and the pathogenesis of lung cancer. The intensity of PPARγ expression indicates the differentiated level of lung cancer, which may be one of markers for lung cancer prognosis.
