摘    要:目的:探讨Neuropilin-1(Nrp-1,神经菌毛素)在小鼠黑色素瘤细胞体外增殖中的作用.方法:用活细胞荧光染料羧基荧光素乙酰乙酸琥珀酰亚胺酯(carboxyfluorescein diacetate succinimidyl ester,CFDA-SE)标记小鼠B16-F10黑色素瘤细胞后,将肿瘤细胞分别用Nrp-1阻断抗体及Nrp-1配体血管内皮生长因子165(vascular endothelial growth factor165,VEGF165)阻断抗体预孵育,体外培养72小时后流式细胞术结合Modfit软件分析肿瘤细胞增殖情况.结果:CFDA-SE标记黑色素瘤细胞生长良好,非抗体阻断组的肿瘤细胞增殖指数显著高于Nrp-1抗体阻断组[(20.67±1.16)% vs (12.67±0.28)%,P<0.01]及VEGF-165抗体阻断组[(20.67±1.16)% vs (15.15±0.83)%,P<0.01],两个抗体阻断组的增殖指数差异也非常显著 (P<0.01).结论:Nrp-1及其配体VDGF-165对小鼠黑色素瘤细胞的增殖有显著地促进作用,阻断Nrp-1产生的抑制肿瘤细胞增殖作用比阻断VEGF165更强;VEGF165/Nrp1直接作用及Nrp-1/Nrp-1直接作用也可能是肿瘤细胞增殖的重要机制.
