骨髓间质干细胞定向分化的实验研究

摘    要:为了探讨体外定向诱导人骨髓间质干细胞(MSC)分化为神经元样细胞的机制,本研究将分离的人MSC进行体外扩增培养,并观察脑心舒定向诱导MSC分化为类神经元样细胞的效应。在光镜下观察细胞形态,用免疫细胞化学法检测神经细胞特异性抗原标志。结果显示人MSC可通过贴壁法成功分离并可在体外大量扩增。脑心舒诱导120min后大部分MSC转变为神经元样细胞,出现胞体和突起,免疫细胞化学染色NSE、nestin呈阳性和GFAP阴性。上述结果提示人骨髓间质干细胞可在体外诱导分化为神经元样细胞。

Abstract:To investigate mechanism umderlying the differentiation from human mesenchymal stem cells (MSCs) to neuron-like cells, MSCs were obtained and expended in culture medium, the effect of the capacity of differentiation MSCs with Naoxinshu which acted as neuron-like cells inducer were performed. Neuron-specific enolase (NSE), nestin, gliall fibrillary acidic protein(GFAP) were detected by immunocytochemistry. The results showed that human MSCs could be expended as undifferentiated cells in culture for more than 20 passages. Simple differentiation method with Naoxinshu which can generate neuron-like cells was achieved within 120 minutes. The expression of NSE and nestin in the neuron-like cells were detected, whereas GFAP was negative in these cells. Our results in these that MSCs could be induced to differentiate into neuron-like cells with Naoxinshu.
