摘    要:研究猪苓多糖对小鼠骨髓树突状细胞(DC)表型及功能成熟的影响。从小鼠骨髓中分离单个核细胞(MNC),用rmGM-CSF、IL-4培养5 d后,实验组加入猪苓多糖(50μg/ml),空白对照组加等量RPMI 1640,阳性对照组加LPS(1μg/ml),3组分别同时作用48 h。扫描电镜观察其体外培养过程中的形态特征,流式细胞仪(FACS)分析其表型特征,混合淋巴细胞反应(MLR)检测其功能。用猪苓多糖刺激的小鼠骨髓MNC具典型的DC形态;细胞表达I-A/I-E、CD11c和分泌IL-12能力比空白对照组增加,具有极强的激发MLR的能力,显示成熟DC的特征。猪苓多糖可以促进体外培养的小鼠骨髓DC的成熟,促进DC诱导的免疫应答启动。

Polyporus polysaccharide stimulates and induces the functional maturation of dendritic cells derived from murine bone marrow
Abstract:The influence of polyporus polysaccharide(PPS) on the phenotypes and functional maturation of dendritic cells(DC) derived from murine bone marrow was investigated in the present study,in which mononucleated cells(MNC) isolated from murine bone marrow were cultivated in RPMI medium 1640 containing rmGM-CSF and IL-4 for 5 days and 50 μg/ml of PPS was then added,thus providing to be used as the experimental groups,while in the blank control group,equal amount of RPMI 1640 medium was added.And in the positive control group,LPS(1 μg/ml) was added.After 48 hours cultivation,the morphologic characteristics of these 3 groups of cells during in vitro cultivation were observed with scanning electron microscopy;phenotypic features were analyzed by FACS,and their immune function was assayed by means of mixed lymphocyte reaction(MLR).It was demonstrated that the MNC from murine bone marrow stimulated with PPS showed the typical features of DC with enhancing abilities to express I-A/I-E and CD11c molecules and secrete more cytokine IL-12 than the controls.In addition,they had strong ability to trigger the MLR,All these features indicate that they possess the typical characteristics of mature DC to stimulate and induce the initiation of immune responses.
