多发性硬化患者T淋巴细胞对不同浓度神经髓鞘及其成分的增殖反应性关系探讨

摘    要:为比较不同浓度神经髓鞘组织、神经髓鞘蛋白及其合成多肽作为抗原对多发性硬化 (MS )和正常人的淋巴细胞诱发增殖反应的量效关系。用HLA DR15亚型MS和正常对照外周血单个核细胞 ,体外培养并检测对不同浓度的人神经髓鞘、脱脂神经髓鞘、蛋白脂蛋白 (PLP )、碱性髓鞘蛋白 (MBP )、PLP178 191和MBP87 10 6多肽 6种抗原的T淋巴细胞增殖反应性。结果发现神经髓鞘或脱脂神经髓鞘作为抗原刺激T淋巴细胞增殖反应的最佳浓度为 5 0~ 15 0 μg/ml,PLP或MBP最佳浓度为 2 5~ 10 0 μg/ml,多肽最佳浓度为 2 0~ 5 0 μg/ml。神经髓鞘抗原刺激T淋巴细胞增殖反应大于单一蛋白分子 ,后者又大于多肽。MS的T细胞对神经髓鞘类抗原有特异性异常增殖反应。

Abstract:To explore the dose response relation of the proliferative responses in T lymphocytes of patients with multiple sclerosis induced with different doses of myelin or its compositions,the proliferative responses of human T cells from mononuclear cells of HLA DR15 type were induced in vitro with 6 peptide antigens including diluted concentrations of intact myelin,delipidated myelin,myelin basic protein (MBP),proteolipid protein (PLP),PLP178 191 and MBP87 106 Experimental results showed that the optimal concentrations for induction of responses induced by myelin and delipidated myelin were 50~150 μg/ml;those induced by PLP or MBP were 25~100 μg/ml,and those induced by various peptides were 20~50 μg/ml The proliferative response induced by myelin protein was stronger than those induced by myelin peptides,and the maximal response was observed in case induced by delipidated myelin with significant difference between T cells of patients with multiple sclerosis and the controls. It concludes that the proliferative responses exist in T lymphocytes of patients with multiple sclerosis induced by myelin antigens
