摘    要:目的:建立快速稳定的人精子核双色荧光原位杂交的实验方法。方法:采用双色荧光原位杂交(fluores cence in situ hybridization, FISH )方法取精子标本用EDTA/PBS液处理,再用二硫苏糖醇(DTT)使精子去凝集,甲醇/冰醋酸固定后制片,变性后与双色荧光直接标记(CEP X、Y)探针杂交。结果:在荧光显微镜下可见标本背景清晰,精子头部染成蓝色,边界清楚,头部有1个绿色荧光信号的精子为 X精子,有 1 个红色荧光信号的精子为 Y精子。精子头部有2个或2个以上信号的精子为染色体数目异常精子。结论:本方法具有快速、稳定、荧光信号强、易分辨等优点,可用于人性染色体非整倍体率的分析研究。

Establishment of An Effective Approach to Dual-color Fluorescence in situ Hybridization of Human Sperm Interphase Nucleus
Abstract:Objective: To establish an effective approach to dual-color fluorescence in situ hybridization of human sperm interphase nucleus.Methods: Dual-color fluorescence in situ hybridization (FISH) was performed on human sperm interphase nucleus using centromeric probes for chromosome X and Y. Results: It was seen under the fluorescence microscope that the sperm showed with one green hybridization signal on the head was X chromosomal sperm, while with one red hybridization signal was Y chromosomal sperm, and those with 2 or >2 fluorescence hybridization signals on the sperm head were abnormal counts for chromosomal sperm. Conclusion: The hybridization signals are visible and distinguishable, and this method is effective. It could be used for analyzing aneupoidy of human sperm chromosome.
