硬腭裸露骨面动物模型的改进及研究

摘    要:目的建立一种能模拟腭裂术后裸露骨面的模型,并进行脱细胞真皮基质的移植,以观察裸露骨面对上颌骨生长发育的影响并检验模型的可行性。方法取3周龄雄性Wistar大鼠36只,随机分为正常对照组;硬腭1/3裸露组;硬腭3/4裸露组。术后12周处死所有动物,测量各组动物硬腭的长度和宽度。结果大体标本观察可见硬腭3/4裸露组裸露骨发生坏死形成腭中缝骨性裂隙,硬腭1/3裸露组伤口为瘢痕组织修复,无死骨形成。硬腭1/3裸露组和硬腭3/4裸露组硬腭宽度均明显小于正常对照组,硬腭3/4裸露组硬腭宽度也明显小于硬腭1/3裸露组。硬腭1/3裸露组、硬腭3/4裸露组及正常对照组硬腭长度虽有差距,但是结果没有统计学显著性。结论腭裂术后上颌骨生长发育障碍是主要由腭部骨性裂隙和裸露骨面上瘢痕形成及收缩造成的。该硬腭裸露动物模型可以模拟腭裂术后硬腭裸露骨面以及硬腭骨性裂隙的情况,并可用于人工及生物材料修复腭裂术后硬腭部裸露骨面的研究。

Abstract:Objective To improve rat model of palate bone denudation and to observe the influence of palate bone denudation on the development of maxillary.Methods Thirty-six three-week-old Wister male rats were randomly divided into 3 groups, control group (group 1), 1/3 palate bone denudation group (group 2), and 3/4 palate bone denudation group (group 3). More 4 three-week-old Wister male rats were used for the feasibility of our model by transplant acellular dermal matrix (ADM) under its palatal mucoperiosteum.Results In group 3, necrotic bone appeared in the mid palate, and all the rats appear osteal cranny. In group 2, there was no necrotic bone, and the wound were repaired by scar tissues. The width of the palate in group 3 and group 2 was significantly narrower than that of group 1. The width of the palate in group 3 was also significantly narrower than that of group 2. There is no difference for the length of the palate in the three groups.Conclusion The osteal cranny in the mid palate and the formation of scar tissue may be the two major factors related to the following maxillary growth deformity in cleft palate patients who received palatoplasty.
