小鼠卵巢组织的深低温保存和移植研究

摘    要:①目的 采用深低温保存方法冷冻保存小鼠的卵巢组织,观察移植后卵巢功能和组织学的恢复情况。②方法 36只昆明种小鼠随机分为3组:假手术组、对照组(新鲜移植组)和深低温保存组。假手术组不切除卵巢,对照组切除卵巢后即行自体原位移植,深低温保存组则将双侧卵巢完全摘除后,行二期手术自体原位移植深低温保存的卵巢。观察所有小鼠的动情周期的变化,并对卵巢的卵泡发育情况进行组织学检查。③结果 假手术组小鼠的动情周期没有任何变化,对照组小鼠的动情周期也在手术后的不同时间内恢复正常,深低温保存组小鼠在完全切除卵巢后,完全失去了动情周期的变化;冷冻复温卵巢自体移植后,多数小鼠的动情周期恢复正常。组织学分析显示,深低温保存卵巢在自体移植后,卵巢可以存活,并有各级卵泡发育。④结论 采用深低温保存方法保存后的小鼠卵巢复温后保持组织活性,自体移植后小鼠的动情周期恢复,卵巢中有正常卵泡发育。

Abstract:Objective To assess the physiological function and histology of the cryopreserved ovarian tissue after autologous transplantation in mice. Methods Thirty-six adult mice were randomly divided into three groups: sham operation group, control group (fresh autograft group) and cryopreservation group, with 12 mice in each group. In the sham operation group, the ovaries were not removed. In the control group, the ovarian tissue was autografted without the freezing-thawing procedure, while the ovarian tissue was autografted after cryopreservation in the cryopreservation group. The estrum of the mice was observed before and after transplantation, and the follicular development of the fresh and frozen-thawed ovarian tissues were also analyzed. Results In the sham operation group, no changes were observed in the estrum and follicular development. All the mice in control group showed normal estrum after operation, the mice in the cryopreservation group resumed estrum after autograft at different time, and the ovaries developed normal follicles. The histology showed that the cryopreserved ovaries which were autografted survived and resumed follicular development. Conclusion The mouse ovaries can survive the freezing-thawing process, and reestablish the estrum and normal follicular development after autologous transplantation.
