摘    要:补骨脂素(psoralen,PSO)是中药补骨脂中的主要成分.研究表明自中药补骨脂中提取的PSO加长波紫外线(UVA),即PUVA对人白血病细胞NB4>、K562>、HL-60的生长均有抑制作用1-2]>,并可诱导白血病细胞凋亡.本研究应用析因设计观察PUVA对NB1>细胞线粒体膜电位(△Ψm)的影响,以探讨PUVA诱导其凋亡的作用机制.

Abstract:Objective: To study the effects of psoralen(PSO) extracted from traditional Chinese medicine psoralea and long wave ultraviolet A(PUVA) inducing the apoptosis on human leukemia NB4 cells. Methods: The factorial design was applied. NB4 cells were taken as the objects and treated with PSO in different concentrations and UVA with 360 nm wl in different time. The changes of ultrastructure of cells were observed under the electron microscope. The apoptosis ratios and the changes of mitochondrial membrane potential were detected through the flow cytometry. Results: 1. PSO plus UVA (PUVA) induced the apoptosis of NB4 cells and the apoptosis ratios showed a dose- and time- dependent manner with the interaction among each factor. 2. Observed under the electron microscope, NB4 cells treated with PUVA had obvious ultrastructure changes with apoptosis. 3.PUVA decreased the mitochondrial membrane potential in a dose -and time- dependent manner with the interaction among each factor. Conclusions: PUVA can decrease the mitochondrial membrane potential and then induce the apoptosis of NB4 cells.
