神经母细胞瘤细胞周期调控基因p21的表达检测

目的　探讨ｐ２１ 基因表达对神经母细胞瘤（ＮＢ） 发生发展的影响。方法　应用酶标链霉亲合素生物素（ＬＳＡＢ）免疫组化技术对２０ 例ＮＢ检测。结果　１８ 例（９０ ％） 可见ｐ２１ 表达阳性细胞，表达指数（ＥＩ）０ ％ ～４５．６ ％ 。有丝分裂核碎裂指数（ＭＫＩ）、瘤体直径与ＥＩ呈负相关，存活时间与ＥＩ呈正相关（ｒ 分别为－０ ．８９ 、－ ０．４９、０ ．４７）。ＥＩ＞２０％ 者ＭＫＩ和瘤体直径均显著低于或小于ＥＩ＜ ２０％ 者（ Ｐ＜ ０．０１），ＥＩ＞ ２０ ％ 者存活时间显著长于ＥＩ＜ ２０ ％ 者（Ｐ＜ ０．０５） 。结论　ｐ２１ 表达水平与之能否抑制ＮＢ细胞周期演进关系密切，表达水平高时，ＮＢ 细胞增殖活性受抑，ＭＫＩ低，预后好；反之，细胞周期演进快，ＭＫＩ高，预后差。

Detection of expression of p21 gene in neuroblastoma

Objective To investigate the role of p21 gene expression in development of neuroblastoma (NB). Methods Expression of p21 gene was detected by using LSAB immunohistochemistry technique in 20 cases of NB. Results Expression of p21 was detectable in 18 out of 20 specimens. The expression index (EI) ranged from 0 to 45.6 %. There were negative correlation between mitotic karyorrhexis index (MKI), tumor diameter and EI (r=-0.89, -0.49, respectively). There was a positive correlation between survival time and EI (r=0.47). The MKI and tumor diameter were significantly decreased when EI>20 % as compared with those when EI<20 % (both P <0.01). The survival time was significantly increased when EI>20 % than that when EI<20 % ( P <0.05). Conclusion The expression level of p21 gene might be critical for its negative regulation of cell cycle progression on the development of NB.;
