脑内移植GFP基因修饰细胞的研究

摘    要:目的为确定外源GFP基因能否作为标记基因来追踪细胞在植入脑内后的生长情况。方法构建GFP基因真核表达质粒,导入PA317细胞,筛选出GFP高表达的PA317细胞克隆,移植入SD大鼠纹状体,作相应部位的组织冰冻切片,用荧光显微镜观察GFP在脑内的表达。结果GFP高表达的PA317细胞克隆,发出强绿色荧光,被移植入SD大鼠纹状体后.仍发出绿色荧光。结论GFP基因可以作为标记基因追踪细胞在植入脑内后的生长情况。

Abstract:Objective To study whether the exogenetic GFP gene can he used as a marker to trace the growth of the GFP gene-modified cells which was transplanted into brain. Method GFP gene eukaryotic express plasmids was constructed and transfected into PA317 cells. The PA317 cells much highly expressing GFP were screened and implanted into the striatum of the SD rats. Brain cryostat sections were prepared and the expression of GFP in the brain was observed using fluorescent microscopy. Result PA317 cells which highly expressed GFP showed green fluorescence. When PA317 cells were implanted into brain striatum of the SD rats, the green fluorescence was also observed. Conclusion GFP gene call be used as a marker to observe the growth of the donor cells implanted into brain.
