摘    要:目的探讨FK506和供体特异性输血在大鼠异位心脏移植中的作用。方法利用大鼠异位心脏移植模型以了解在移植当天或移植术后第4日进行供者特异性输注(DST)的基础上,应用FK506能否延长移植物的存活。结果在移植当天行DST或单独用FK5061mg/kg连续10天,可将移植心中位存活时间从对照组的5天分别有效延长至7天和11天,而FK506与DST联合应用时并不产生增强效应。结论FK506和DST单独应用时虽均能延长大鼠同种移植心存活,但是它们没有协同作用。

Abstract:Objective To explore the role of donor specific blood transfusion (DST) and FK506 on heterotopic cardiac transplantation in rats. Method DST was given on the day of transplantation or on the posttransplant day 4 in a model of rat heterotopic cardiac transplantantion. FK506 was used to observe the survival of the grafts. Result DST 2 ml on the day of transplantation or FK506 1 mg/kg for 10 days alone can effectively prolong the allograft survival from 5 days in the control group to 7 days of DST and 11. 5 days of FK506 (P = 0.0040, P =0. 0020 versus control, respectively). FK506 did not enhance the beneficial effects of DST on allograft survival (P = 0. 1248 FK506 1 mg/kg 10 days course plus DST group versus FK506 only, P = 0. 2948 FK506 1 mg/kg for 10 days course plus DST on day 0 and 4 group versus FK506 1 mg/kg for 10 days course plus DST on day 0) although FK506 prolonged the graft survival significantly (P = 0.0050 FK506 1 mg/kg for 10 days course plus DST on day 0 group versus DST on day 0 only, P = 0. 0163 FK506 2 mg/kg for 10 days course plus DST on day 0 group versus FK506 1 mg/kg for 10 days course plus DST on day 0). Conclusion Both DST and FK506 alone can prolong the allograft survival in rat cardiac transplant model significantly, but FK506 and DST did not prolong the graft survival in a synergistic manner.
