摘    要:目的 探讨睡眠呼吸暂停模式间歇低氧(IH)对大鼠外周血淋巴细胞亚群凋亡的影响.方法 将16只雄性Wistar大鼠随机分为常氧对照组(NC,n=8)和间歇低氧(IH)4周组(IH4,n=8),IH暴露环境最低氧浓度为5%,IH频率为30次/h.暴露完成后取外周血分离淋巴细胞,流式细胞术检测淋巴细胞亚群凋亡情况.结果 和NC组比较,IH组CD4 T细胞凋亡率(15.17±1.43)%低于NC组(17.97±1.76)%,CD8 T细胞凋亡率(17.23±1.07)%低于NC组(19.7l±1.93)%,NK细胞凋亡率(22.07±1.45)%高于NC组(17.33±1.04)%,B细胞凋亡率(17.88±1.85)%高于NC组(13.94±1.76)%(P均<0.01).结论 IH暴露改变大鼠外周血淋巴细胞亚群的凋亡状态,导致免疫功能异常,可能加重内皮细胞功能障碍.

Abstract:Objective To examine the effect of intermittent hypoxia (IH) on lymphocyte subset apoptosis in a mouse model of sleep apnea.Methods Sixteen male Wistar rats were randomly divided into 2groups (n =8 each):normal oxygen control group (NC) and intermittent hypoxia for successive 4 weeks group (IH4).For IH expose,the minimum oxygen concentrations was 5% with an apnea hypopnea index of 30/h.Apoptosis of lymphocyte subset was detected using flow cytometry assay.Results Compared with the NC group,apoptosis of CD4 and CD8 T cells was decreased significantly in the IH4 group (P < 0.01),while apoptosis of B and NK cells increased significantly in the IH4 group (P < 0.01).Conclusion Intermittent hypoxia regulates differentially apoptosis in circulating blood lymphocyte subsets,which may lead to altered immunologic function and contribute to endothelial dysfunction.
