摘    要:目的 :探讨黄芪在缺血再灌注损伤早期肿瘤坏死因子 (TNF)介导的心肌细胞凋亡中的作用。方法 :将大鼠分为对照组、实验组和黄芪组 3组。采用大鼠缺血再灌注离体心脏模型 ,检测心肌组织TNF含量与细胞凋亡情况。结果 :(1)实验组心肌组织中的TNF水平较对照组明显升高 (P <0 .0 5 ) ,黄芪组与实验组相比则明显降低 (P <0 .0 5 )。 (2 )对照组透射电镜 (TEM )未见发生凋亡的心肌细胞 ,也未见TUNEL阳性细胞 ;实验组和黄芪组 ,TEM可见心肌组织中有趋于凋亡的心肌细胞 ,并可见少量TUNEL阳性细胞。结论 :在缺血再灌注损伤早期黄芪可以明显降低心肌内TNF水平 ,但未见其对细胞凋亡有直接作用

Abstract:AIM: To observe the effects of Astragalus membranaceus on apoptosis-induced by tumor necrosis factor (TNF) in early ischemia reperfusion injury. METHODS: The Wistar rats were divided into three groups randomly, control group, experiment group and Astragalus membranaceus group. Level of myocardial TNF and cardiac myocyte appoptosis in reperfused isolated heart model in rats were examined. RESULTS: The level of myocardial TNF in experiment group was significantly elevated compared with control group (P<0.05), and that of Astragalus membranaceus group was significantly decreased compared with experiment group (P<0.05). Apoptosis in cardiomyocytes was not observed by TEM and TUNEL in control group. In contrast, a few myocytes appearing early changes of apoptosis and TUNEL positive cells were observed in experiment or Astragalus membranaceus group. CONCLUSION: In early ischemia reperfusion injury, Astragalus membranaceus may significantly decrease the level of myocardial TNF but has no direct effect on appoptosis in cardiomyocytes.
