[Ca2+]i与阻塞性黄疸大鼠肾系膜细胞损伤的研究

摘    要:目的 探讨阻塞性黄疸大鼠胞浆钙活性的变化及其致肾损伤的作用机制.方法 60只SD大鼠,随机分为正常对照组、假手术组、实验组(再分为胆总管结扎后15 d和21 d两个亚组),每组15只.胆总管双重结扎法建立阻塞性黄疸大鼠模型,切取肾脏行肾小球系膜细胞原代培养,激光共聚焦显微镜法测定细胞胞浆钙离子浓度.结果 肾小球系膜细胞胞浆钙离子浓度明显升高(P<0.05).结论 TNFa致阻塞性黄疸大鼠肾脏系膜细胞线粒体损伤中细胞浆内钙离子超载起着重要的作用.

Abstract:Objective To explore the effect of the biological activity of calcium of the cultured glomerular mesangial cells(GMCs) on the kidney injury of rats with obstructive jaundice and the underlying mechanism. Methods Forty-five male sprague-dowley rats, weighting 200 to 250 g were-randomly assigned to two groups:control-group (sham-operation)and experiment group. Except the control group, the experiment group were rendered jaundiced by doubly ligating the common bile duct. Clomerulus were dissected from the animal and used to set up the primary culture of CMCs. The concentration of calcium were measured by confocal microscope. Result The concentration of calcium were increased (P< 0.05) in the rats with obstructive jaundice compared to the controlled. Conclusion Changes of concentration of the calcium has significant effect on the kidney injury of the rats with obstructive jaundice.
