Simultaneous determination of seven active ingredients in Chuanxiong Chatiao Granules by HPLC-CAD

OBJECTIVE To develop an HPLC-CAD method for simultaneous determination of seven active ingredients of ferulic acid,glycyrrhizic acid,coniferly ferulate,senkyunolide A,Z-ligustilide,imperatorin and isoimperatorin in Chuanxiong Chatiao Granules. METHODS Chromatographic separation was performed on an Agilent Eclipse XDB-C₁₈(250 mm×4.6 mm,5 μm) column with 10 mmol·L^-1 ammonium formate(A) - 80% acetonitrile(B) as the mobile phase for gradient elution,flow rate of 1.0 mL·min^-1,column temperature of 35℃,nebulization temperature of detector 35℃,acquisition frequency 10 Hz and filtration constant 5.0. RESULTS Ferulic acid,glycyrrhizic acid,coniferly ferulate,senkyunolide A,Z-ligustilide,imperatorin and isoimperatorin were linear in the ranges of 0.55-11.00 μg·mL^-1(r=0.999 3),5.90-118.00 μg·mL^-1(r=0.999 4),2.51-50.20 μg·mL^-1(r=0.999 5),5.02-100.40 μg·mL^-1(r=0.999 4),4.99-99.80 μg·mL^-1(r=0.999 8),1.03-20.60 μg·mL^-1(r=0.999 5) and 0.59-11.80 μg·mL^-1(r=0.999 5) respectively;The average recoveries were 97.78%,97.76%,97.54%,98.09%,97.98%,97.89% and 98.05% and RSDs 0.96%,1.50%,0.84%,1.49%,1.50%,1.37% and 1.65% respectively.Precision,reproducibility and stability of solution were in accord with the regulations.The content ranges of seven ingredients were 0.45-0.60,4.93-6.20,2.01-2.73,2.63-3.43,4.72-5.70,1.13-1.84 and 0.38-0.52 mg/bag. CONCLUSION The above method is accurate,sensitive,convenient and reproducible for quality control of multicomponents in Chuanxiong Chatiao Granules.
